Postdoctoral Position and PhD Student Position. Within the department of Physiology, section Cell Physiology, of the Radboud University Nijmegen Medical Center, we are seeking a Post-doc and a PhD student. The section Cell Physiology has a long-standing tradition in transport physiology in epithelia and build up a substantial know-how and infrastructure in the field of molecular and cellular physiology of water and electrolyte transport. The department is, together with basic science and research-oriented clinical departments of the Radboud University Nijmegen, housed in the Nijmegen Center of Molecular Life Sciences (NCLMS). This new setting has created a unique scientific infrastructure allowing stimulating and innovative science. The Ca2+ and Mg2+ reabsorbing activity of the kidney determine the net excretion of these divalent ions for the entire body and, therefore, the body Ca2+ and Mg2+ balance. The maintenance of this balance is of crucial importance for many vital physiological functions including neuronal excitability, muscle contraction and bone formation. In healthy adults, the renal excretion of Ca2+ and Mg2+ is critically balanced by gastrointestinal absorption. A major breakthrough in completing the molecular details of the above-mentioned pathways was the recent identification of the Transient Receptor Potential channel (TRP) superfamily. Our group identified the epithelial Ca2+ channel family that consists of two members TRPV5 and TRPV6. Recently, another TRP channel (TRPM6) was postulated as the renal Mg2+ influx mechanism. The epithelial Ca2+ channels could be involved in the multifactorial pathogenesis of disorders ranging from idiopathic hypercalciuria, stone disease and postmenopausal osteoporosis, whereas TRPM6 was mutated in patients with hypomagnesemia with secondary hypocalcemia (HSH). TRPV5, TRPV6 and TRPM6 display the defining properties for being the gatekeepers in active Ca2+ and Mg2+ reabsorption and are prime targets for hormonal control of Ca2+ and Mg2+ flux from the urine space to the blood compartment. Our recent studies substantiate the gatekeeper function of TRPV5 by showing that TRPV5 knockout mice display a robust hypercalciuria causing a compensatory Ca2+ hyperabsorption and disturbed bone formation. Understanding the molecular mechanism by which idiopathic hypercalciuria and HSH occurs, would allow clinical investigators to screen families with the disease before the onset of the overt disease and possibly direct treatment at the specific underlying molecular defects. Projects aim, therefore, to unravel the mechanism of Ca2+ and Mg2+ (re)absorption to provide insight in the molecular basis of these diseases. Requirements: Postdoc (2-3 years): PhD in biomedical sciences, experience with molecular biology; permission to work with animals is a preference (art. 9). Salary for Post-doc will be dependent on the qualifications and experience from Euro 2625,-  up to a maximum of Euro 4027,-. (gross amount). PhD student (3-4 years): finished university education in (medical) biology or chemistry (masters or drs.); experience with molecular biology; permission to work with animals is a preference (art. 9). Salary for PhD from Euro 2179,- (gross amount). Information and Application: Further information can be obtained from Dr. J. Hoenderop (+31-24-3610571) or Prof. Dr. R. Bindels (+31-24-3614211).
